Patapoutian en visite à Strasbourg https://neurostra.unistra.fr/actualites/actualite/news/a-patapoutian-en-visite-a-strasbourg/ Nous avons eu le plaisir d’accueillir à Strasbourg Ardem Patapoutian, Prix Nobel de physiologie ou médecine 2021 et CNRS Fellow-Ambassadeur  !

Ardem Patapoutian, professeur au Scripps Research Institute en Californie, est Prix Nobel de physiologie ou médecine 2021 pour la découverte des récepteurs impliqués dans la mécanosensation, à l’origine du toucher et de la proprioception. Nous avons eu l’immense joie de l’accueillir pour 3 jours sur Strasbourg dans le cadre du programme « CNRS fellows-ambassadeurs ». Lors de cette visite, des échanges riches ont eu lieu avec les étudiants, doctorants de l’Ecole Universitaire de Recherche Euridol et de l’lnstitut Thématique (ITI) NeuroStra, ainsi qu’avec les équipes et chercheurs locaux en neurosciences et les représentants du CNRS et de l’Université de Strasbourg. Au-delà de ces rencontres scientifiques, des moments de convivialité ont aussi permis de lui faire découvrir Strasbourg et l’Alsace. Le lundi 26 mai, plus de 160 personnes ont pu assister à sa conférence "May the Force be with you! Piezo channels in mechanosensory biology", distinguished lecture USIAS (University of Strasbourg Institute for Advanced Science).

Collaborative work involving 2 clinical departemnt at HUS (opthalmology and pediatrci neuro-oncology)

International collaboration with European consortium (8 clinical sites in 7 countries: Strasbourg, Milan, Padova, Barcelona, Vienna, Utrecht, London, Copenhagen).


Pre-requisite

Master in Neurosciences or equivalent (MSc, MD). Comprehensive knowledge of the visual system.

Minimum B2 level in English. Knowledge of another european language is a plus. Comprehensive knowledge of Python (coding), biomedical statistics/epidemiology. Strong interest for innovating technologies. Knowledge of clinical trials regulations.

To survive in a rapidly dynamic world, the brain predicts the “what”, “where” and “when” of upcoming events and proactively adjusts perception, attention and action. The cerebellum has traditionally been associated with prediction and timing in the motor domain, while proactive perception and attention were associated with oscillatory mechanisms in cortical circuits. I will present a series of studies that combined computational modelling, psychophysics, and EEG recording in healthy and neurological populations to investigate mechanisms of dynamic prediction and the potential role of the cerebellum beyond motor control. These reveal that the cerebellum is indeed involved in non-motor prediction, of the timing of sensory events, as well as of semantic content in language. This is then leveraged to uncover causal cerebellar control of temporal variability in cortical circuits, identify unique entrainment mechanisms to isochronous streams, and study the computational overlap between implicit and explicit timing. Altogether, these findings expand our understanding of subcortical-cortical interactions in supporting prediction, timing, and high-level cognition, and inspire translational applications.

news-4861 Fri, 14 Feb 2025 10:40:27 +0100 Synaptic plasticity in the hippocampus in health and disease https://neurostra.unistra.fr/evenements/tous-les-evenements/news/synaptic-plasticity-in-the-hippocampus-in-health-and-disease/ Long-term potentiation (LTP) is widely recognized as a fundamental property of most synapses in the mammalian brain, with NMDA receptor-dependent LTP (NMDAR-LTP) being the predominant form. LTP remains the most intensely studied and best-understood mechanism of synaptic plasticity, with profound implications for learning, memory, aging, and neurological disorders. I will summarize the early work leading up to the understanding of the molecular basis of LTP in the CA1 region of the rodent hippocampus. Next I will summarize work that supports the view that Alzheimer’s disease is principally a dysregulation of synaptic plasticity. Lastly I will summarize two on-going, unpublished studies: 1. The effects of eliminating tau on synaptic plasticity in rats. 2. Alterations in striatal synaptic plasticity in a mouse model of autism that lacks co-morbidities.

    We study the contribution of microglial cells and underlying mechanisms of neuronal and glial dyshomeostasis in neurodegenerative conditions including Alzheimer´s disease (AD) and the childhood dementia Niemann-Pick type C (NPC). We use translational approaches to identify molecular fingerprints of microglial dysfunction and identify novel cellular targets to target and monitor neuroinflammation across neurodegenerative disorders.

    Our research aims to explore the intricate interactions between Müller cells, microglia and neurons in both healthy and diseased retinas. We are particularly interested in understanding the effects of AAV-vectored gene therapies targeting these interactions, in particular the coordination of the retinal immune response, for example by modulating retinal complement homeostasis or glucocorticoid receptor signaling. We demonstrate that our approach of delivering therapeutic gene expression to Müller cells can improve disease progression in models of diabetic retinopathy or ischemic stress, independent of the underlying cause of the disease, and thus may benefit patients who are not candidates for currently available treatment strategies, since most, if not all, retinal diseases involve Müller cell gliosis and inflammatory processes that potentially accelerate neuronal dysfunction.
