The ability to tell time, represent and encode temporal information, and use past experiences to predict the future, are among the most fundamental computations the brain performs. But how do networks of neurons tell time and encode temporal information in a flexible manner? Precisely because of the importance of time, we argue that most neural circuits are capable of temporal computations. But in contrast to the clocks on our wrists that rely on the same mechanism to tell time across a wide range of scales, the brain engages many different mechanisms and circuits to tell time. We and others have proposed that on the scale of seconds timing relies on neural population clocks: time-varying patterns of neural activity that emerge from the dynamics of recurrent neural circuits. More generally, we propose that such dynamic regimes represent a fundamental computational strategy in the brain.

    We study the contribution of microglial cells and underlying mechanisms of neuronal and glial dyshomeostasis in neurodegenerative conditions including Alzheimer´s disease (AD) and the childhood dementia Niemann-Pick type C (NPC). We use translational approaches to identify molecular fingerprints of microglial dysfunction and identify novel cellular targets to target and monitor neuroinflammation across neurodegenerative disorders.

    Our research aims to explore the intricate interactions between Müller cells, microglia and neurons in both healthy and diseased retinas. We are particularly interested in understanding the effects of AAV-vectored gene therapies targeting these interactions, in particular the coordination of the retinal immune response, for example by modulating retinal complement homeostasis or glucocorticoid receptor signaling. We demonstrate that our approach of delivering therapeutic gene expression to Müller cells can improve disease progression in models of diabetic retinopathy or ischemic stress, independent of the underlying cause of the disease, and thus may benefit patients who are not candidates for currently available treatment strategies, since most, if not all, retinal diseases involve Müller cell gliosis and inflammatory processes that potentially accelerate neuronal dysfunction.

    Téléthon charity night Feedbacks – 19.12.2023

    The last tuesday before Xmas holidays, we organized a charity concert at the K'fet des Sciences, with music played by the Mygales Détendues and collected money was given to Téléthon.

    Thanks to you and your amazing energy, it was a huge success and we collected 400€ that were distributed to Téléthon.

    Doctoneuro's sweaters are on their way !

    We do not forget you, the warmy 2023-2024 doctoneuro sweater that you ordered are on their way to join your labs and classrooms.

Nevertheless, surprisingly little is known about how long-lasting memories are formed and stored. Over the past decade, engram cells have emerged as a plausible physiological substrate of the memory trace, but the molecular mechanisms therein to encode memories remain incompletely understood. Here, we combine c-Fos driven engram tagging technologies with in vivo epigenetic editing tools to assess whether and the extent to which epigenetic modifications within the memory trace cells contribute to memory encoding. Here we show that epigenetic editing of a single site on the genome – using CRISPR-dCas9 coupled to transcriptional activators or repressors – is sufficient to alter memory performances. Engram-specific, dCas9-VPR mediated upregulation of a master regulator of synaptic plasticity improves recent fear memory, whilst expression of dCas9-KRAB-MeCP2 in the same cells at the same locus impairs fear memory recall. Overall, these findings show how the modulation of epigenetic marks in a cell type and locus-specific manner can impact neuronal function and memory capacity; at the same time, they deepen our understanding of the molecular mechanisms of traumatic memory storage.
